Depends on the bot.
You can use autoit bots that do nothing with memory and really only respond to what is on the screen.
Some bots even autoit ones read memory to create actions.
So we could say both methods are possible right?

hardcore memory reading and writing to make a fast bot tho. when u purchased a bot like this u want just profit as fast as possible. u will get a ban in the near future, most likely u ve got enogh time to make enogh money so its okay. never bot on ur main! u will receive a ban in every case. just a matter of time. and if u stop botting u are still bot save. a game master just need to look up ur old stats and u are banned anyway. so never on ur main.
There are three "maincategories" of D3 bots: Memory reading, Pixel reading, Memory reading + writing/injection
Both Immortalbot, Demonbuddy and Hellbuddy are using good methods to memory read values. Basically all big public bots you come across will use memory reading of some sort, and some will use injections or memory edits along with the memory reading. I think all three of these bots use injection of some kind, but it doesn't seem that Warden scans those specific values at this time.
They use this technique because creating a pure memory reading bot is a difficult task to perform, at least if you're looking to create a bot similar to these. One example of such bot would be Shadowbot for WoW, which took year(s) for skilled coders to develop.
Almost all of the AutoIt scripts on this forum are using pixelreading techniques only, which means they scan the screen for certain pixels or images, and respond to what it finds, without reading or writing anything to Diablo's memory.
I hope I didn't say anything that was false information, and perhaps shed some light on OP's question.
