No. You can use it whenever you want (its like u are invulnerable but u have to press the button )

Do you mean the trinket or just the item? For the item use the modified cache :P
Oh and for changing all the values just drop them all down into the lower window and highlight them all. Then just doubleclick on one of those values (maybe STRG+ Doubleclick :P)
