Act 1 Quest - Rumford at the Gate.

Killing 3 Wretched Mothers gives 16k experience so you only need to hit 3 of them on your way to the Queen.
Killing the Queen and turning in gives 44k experience.

Throw on some movement speed bonuses and youre set.

dont forget to go to the "dank cellar" (sometimes open) for Sarkoth, he drops yellows most of the time

This is actually quite effective, at least it's probably the best way to level fast before the nerf of Azmodan trick.
I leveled 59 half way to 60 in about 15 minites.

Just to expand on this. If you follow through with the next step (legacy of cain) where you and leah go to her mothers hut you can get another 16500exp/600 gold and a yellow mob to kill. It doesnt add much more time.

i was able to do 150k with some mobs on the way. I dropped traps as a DH
