Depends on what you are attempting to do, pvp or level/ raid.

    PvP:

I suggest you watch this video on what to do for pvp UI. Though it is long, it has the main Addons used for pvping. He covers everything. I strongly suggest you watch it.

    http://warcraftmovies.com/movieview.php?id=89725

    Leveling:
I dont know what to tell you here. There are plenty of great leveling Addons such as QuestHelper, and Carbonite. When i level, i prefer the Zygor's 1-80 Horde/Alliance Ingame Guide. It can be found in the WoW Guides section.

Feel free to search Here for any other helpful leveling addons.

    Raiding:

If you don't like a crouded screen, i suggest you stay out of here. Most UI compilations I've seen are based on raiding. If you are at all interested in these, please go Here for those UI's.

Well, I hope this helped you

Addons are a lot personal preference over necessity. That being stated, there are some incredibly useful addons out there. Most, if not all, of the following can be found on WoWInterface

If you do any AH'ing at all, Auctioneer is a godsend for posting/pricing stuff. It has a LOT of very powerful features, and if you take time to learn it, it will help you make big money on the AH.

If you group a lot, you will want Omen for a threat meter, so you know when to stop DPS, or when you as a tank aren't putting out enough threat.

I personally am in love with addons that let me move my action bars around, or completely replace them. Bartender 4 used to be the king of this, but is having some current difficulties with vehicles. Dominoes does it well, and has all the same features as far as I can tell. CTBar from CTMod - A World of Warcraft Mod is good too.

I *highly* suggest using Carbonite Quest or (if they get/keep it hacked here) the full Carbonite for levelling/questing. It has a phenominal Google-like map and fairly intuitive quest help markers.

If you're not using Carbonite, I suggest getting these three addons as a set. Cartographer from Curse, LightHeaded and DoubleWide from WoWInterface They will help you with questing more than you could imagine.

If you like damage meters of some kind, find Recount on Curse as it is one of the best.

For a unitframe replacement, I *highly* suggest X-Perl from WoWInterface

Couple class specific ones, if you are a paladin, by god, get Pally Power. There is no better blessing management tool anywhere. If you happen to be a tankadin, get Tankadin2 and both it's plug-in's as well. If you play a warlock, I suggest ForteXorcist.

For inventory management, I suggest either Combuctor or TBag by Shefki both from WoWInterface

For helping with Tradeskills, I suggest Skillet, combined with AckisRecipeList.

Some really basic automation of boring tasks, find Automaton. it'll help you get rid of some of the clutter of playing. Autosells greys, autorepairs, auto queue for BG's, auto release on death in BG's, automatically fills in the name of the last person you sent a mail to at the mailbox, automatically skips gossip screens at NPC's

Those are probably the best among the addons I use currently. I wish you a lot of luck and patience in finding the right set up for yourself.
